Abstract

Present study aimed at isolating pure population of blood and milk neutrophils from Karan Fries (KF) cows and to standardize immunolocalisation of surface adhesion molecules. The study employed sensitive gradient based separation technique using two Histopacques for the isolation of both blood and milk neutrophils. After the isolation, cell purity was checked by employing Field’s stain. The cells were used for the immunolocalisation of surface adhesion receptors namely L- Selectin (CD62L) and Integrin Beta (CD11b). Cells were observed by using an inverted epi-flourescence microscope with blue/FITC using FITC as a probe. The results revealed successful labeling of blood and milk neutrophils for CD62L and CD11b.
