Abstract
The present study was designed to determine the relative expression of growth differentiation factor 9 (GDF-9), hyaluronan-synthase-2 (HAS-2) and prostagladin-endoperoxide synthase-2 (PTGS-2) in good (Grade I and II) and poor (Grade III and IV) quality oocytes cumulus cells as biomarkers of oocyte quality. A total 144 caprine ovaries were collected from local slaughterhouse. A total of 391, 344, 218 and 146 oocytes of grades I, II, III and IV, respectively, were recovered by aspiration method with the respective recovery rates of 35.58%, 31.30%, 19.83% and 13.29%. Total RNA was extracted from the oocytes and cumulus cells and real time PCR amplification of GDF-9, HAS-2 and PTGS-2 was done using â-actin and GAPDH as endogenous controls. The poor quality oocytes and CCs were kept as a calibrator group. The relative fold changes of GDF-9 (2.15 folds) in good quality oocytes were significantly (p<0.01) up-regulated as compared to the poor quality oocytes. Similarly, the relative expression of HAS-2 (1.70 folds) and PTGS-2 (4.34 folds) genes in cumulus cells were significantly (p<0.01) up regulated as compared to the poor quality oocytes The current study revealed a significant higher differential expression of GDF-9, HAS-2 and PTGS-2 in good quality COC’s as compared to poor quality COC’s.
