Abstract

The objective was to study the effectiveness of calcium ionophore treatment for activation of oocytes and theirdevelopmental capacity in vitro. A total of 5585 oocytes were retrieved from 1154 caprine ovaries of abattoir origin. Recovery rate of oocytes per ovary was recorded 4.84. Oocytes were matured in TCM-199 containing 10% FBS, sodium pyruvate 0.25 mM, L-glutamine 100 μg /ml, LH 5 μg/ml, FSH 5 μg/ml, BSA 3 mg/ml and gentamicin 50 μg /ml for 27 hours in humidified atmosphere of 5% CO2 at 38.50C in CO2 incubator. After parthenogenetic activation treatment using 5μm calcium ionophore for 5 min, oocytes were treated with 2mM DMAP for 3 hours and then cultured in embryo development media. The results suggested that the overall developmental rate through parthenogenetic activation by 5μm calcium ionophore for 5 min recorded was 56.02% (29.91%, 2-cell; 11.01%, 4-cell; 9.15%, 8-16 cell; 4.94%, morula; 1.01%, blastocysts) in vitro matured caprine oocytes.
