Abstract

Present study was conducted to evaluate oxidative stress and enzymatic activity in response to the addition of different concentration of melatonin in Kankrej bull semen during cryopreservation. A total 24 ejaculates were collected at weekly interval up to eight weeks period from three healthy Kankrej bulls. Each semen ejaculate was divided into four equal aliquots and extended with tris-based extender containing 0 mM (control), 1 mM, 2 mM and 3 mM concentration of melatonin and cryopreserved. Malondialdehyde (MDA), glutathione reductase (GSH), aspartate amino transaminase (AST) and alanine amino transaminase (ALT) were estimated from seminal plasma at post-dilution, post-equilibration and post-thaw stages of cryopreservation. Extender with 2 mM melatonin showed a significant (p<0.05) decrease in MDA, AST and ALT level whereas, GSH level increased significantly (p<0.05) during all the stages of cryopreservation as compared to other concentrations. It was concluded that the addition of melatonin at 2 mM concentration in the tris-based extender gives better protective effect to spermatozoa against oxidative damage compare to dilution without addition of melatonin and dilution with 1 mM and 3 mM concentration of melatonin during cryopreservation.
