Abstract

The present study evaluated the effect of leptin and IGF-I supplementation on in vitro maturation of goat oocytes. Abattoir derived cumulus oocyte complexes were matured in vitro in maturation medium randomly supplemented with either no supplement (group I, control), 20ng/mL leptin (group II) or 100 ng/ mL of insulin-like growth factor-1. The in vitro maturation was assessed on the basis of the first polar body extrusion (nuclear maturation). The matured oocytes were subjected to in vitro fertilization and resultant embryo production was assessed. The nuclear maturation rates did not differ (93.43±1.59, 97.55±0.40 and 94.46±0.91 per cent) between the three groups, respectively. Similarly, there were no significant differences in morula 20.57±1.48, 22.16±1.77, 21.14±1.79% and blastocyst development 10.75±0.31, 12.40±0.90, 11.70±0.30% respectively in all three groups. It was concluded that there was no significant effect of leptin and IGF-I supplementation on in vitro maturation and blastocyst development of goat oocytes.
