Abstract

The present study was conducted to establish the primary culture of mammary epithelial cells in the Bakarwali goat of the Jammu region. The luminal epithelial cells of a Bakarwali goat’s mammary tissues were extracted by mincing and grown in the presence of complete media (DMEM F-12, FBS, ITS and EGF). Differential digestion, selective trypsinization, and continuous passaging were used to isolate the cells. By observing the doubling time of epithelial cells, growth properties were evaluated. Trypan blue exclusion method was used to determine cell viability and number. Specific markers (Krt18, Csn2 and Vim) were assessed by their mRNA expression analysis. The primary culture of goat mammary epithelial cells exhibited typical cobblestone morphology and growth characteristics. The primary mammary epithelial cell culture would facilitate research in mammary gland development and function.
