Abstract
Myostatin (MSTN), also known as growth and differentiation factor 8 (GDF8) is a negative growth regulator of muscle development. The present study aimed to characterize the complete coding sequence (CDS) in Muzaffarnagari sheep breed. In the current study, all three exons of MSTN gene were cloned and characterized in Muzaffarnagari sheep and deduced complete CDS was submitted in NCBI database with Accession No. MT038361. Multiple sequence analysis of Muzaffarnagari sheep MSTN CDS with other breeds and species showed 100% similarity with Indian Nilgiri and other exotic sheep breeds, while 99.9% similarity with Indian Bandur sheep breed at the nucleotide level. At amino acid level, MSTN protein is 99.7% similar to Indian Nilgiri and other exotic sheep breeds and 99.5% similar to Indian Bandur sheep. Further, sequence comparison revealed synonymous nucleotide substitutions at nucleotide positions 126 (T C), 189 (A G) and 903 (C T) between Muzaffarnagari and other Indian and exotic sheep breeds. However, a non-synonymous (A T) nucleotide substitution at position 821 was observed in the Bandur sheep breed that led to the presence of a “V (Valine)” instead of “E (Glutamic acid)” at amino acid position 274. Phylogenetic analysis revealed that the Muzaffarnagri sheep MSTN was more closely related to that of goat, thar, ibex and camel.
