Abstract
Osmanabadi breed of goat from the state of Maharashtra is known for its adaptability, production of good quality chevon, and high prolificacy. Growth rate is one of the most important traits of economic importance in goats Insulin-like Growth Factor Binding Protein-3 (IGFBP-3) and Bone Morphogenetic Protein-4 (BMP-4) genes are known to have a positive association with the growth rate in goats. In the present research work, the blood samples of Osmanabadi goat were subjected to analysis of IGFBP-3 and BMP-4 gene loci following DNA isolation. The PCR-RFLP and PCR-SSCP techniques were used to identify polymorphism of IGFBP- 3 and BMP-4 genes, respectively. The RE digestion of the gel extracted PCR product (654 bp) of IGFBP-3 gene using HaeIII yielded a single restriction pattern of two fragments of sizes 201 bp (appeared as a single band) in all the animals belonging to the Osmanabadi breed under study, However, an expected band of 87, 67 and 57 bp were not visible probably due to their small size. The present findings suggest that none of the animals carries mutant alleles, revealing the absence of polymorphism in the Osmanabadi goats. PCR-SSCP method was used to detect mutations within the amplified PCR products of the BMP-4 gene locus. A similar pattern of the band was observed in all the DNA samples under study. We could not detect any mutation in the intron 2 region of the BMP-4 gene of Osmanabadi goats, indicating that the goats are carrying only wild type and hence no polymorphism was detected.
