Abstract

The present study was designed to investigate the polymorphism of DGAT1 gene (411bp of Exon VIII) and to assess its association with milk yield traits in Gaolao cattle of Maharashtra. Data on daily milk yield, total milk yield, Fat and SNF was collected from unrelated 32 lactating Gaolao cows maintained at Bull Mother Farm, Pohara Dist. Amravati, Maharashtra. A reported primer with Forward primer- 5’-GCACCATCCTCTTCCTCAAG-3’ and Reverse primer- 5’-GGAAGCGCTTTCGGATG-3’ were selected for PCR-RFLP and the amplified fragment was digested using Eae1 restriction enzyme. The Eae1 restriction endonuclease detected a specific restriction pattern in all 32 Gaolao cows indicating uncut fragment indicating band size as 411 bp of DGAT1 gene, which exhibited a monomorphic pattern  (KK) lacking restriction site. The frequency of observed genotypes AA, KK and KA was estimated as 0, 1, and 0 respectively and accordingly, the gene frequency for K allele was estimated as 1 in the studied Gaolao population. The association of the DGAT1 gene with milk traits in Gaolao cattle cannot be investigated due to the observed monomorphic pattern. The overall least squares mean for daily milk yield, total milk yield, Fat and SNF in Gaolao cattle were 1.06±0.15 kg, 195.25±21.0 kg, 3.45±0.13 per cent and 7.34±0.11 per cent, respectively.  The effect of lactation length, parity and season of calving were observed as non-significant sources of variation for all studied milk traits in Gaolao. The present investigation revealed a monomorphic pattern for KK genotype with respect to locus 411 bp of DGAT1 gene in the studied Gaolao population, which was suggesting the complete absence of ‘A allele’ and fixation of ‘K allele’.
