A herd of Sahiwal cattle was studied for determination of genetic polymorphism in intron region of resistin (RETN) gene and its influence on production and reproduction traits of animal. The polymorphism in intron-3 region of bovine RETN gene was determined by PCR-SSCP method. The amplification of gene fragment formed PCR product of 195 bp that was subjected to SSCP for identification of genetic polymorphism. The RETN/SSCP assay revealed two genotypes; homozygous AA and heterozygous AB with 19.30% and 80.70% frequencies, respectively and the allelic frequencies for A and B alleles were 0.596 and 0.404, respectively. The RETN/SSCP genotypes revealed significant association with lactation period (LP), milk yield in 300 days (MY300) and peak yield (PY). RETN/SSCP assay showed longer LP in AB than AA genotype in first lactation. The study also revealed significantly higher MY300 and PY in AB than AA genotype in fourth lactation of Sahiwal cattle. In conclusion, the study suggests that PCR-SSCP assay is competent to detect polymorphism in intron region of RETN gene and its influence on production and reproduction traits indicates that B allele of this gene could be important in marker assisted selection of Sahiwal cattle for improvement in production potential and reproductive performance. However, further studies could be performed to explore this genetic polymorphism in another breed and population of cattle.
